Effects of 5-Fluorouracil on the Growth of Chick Embryo


Abstract
Administration of 5-fluorouracil into various species of animals has widely been reported since last decade, however, none of them were utilized a growth formula proposed by Zimmer man and Huxley, In this paper we attempt to analyze the specific growth rate in general and organ to organ relative growth rate using the chick embryo under the influence of 5-fluorouracil which is well known as an antimethylation. Congenital malformation of chick embryo were not considered.
On the sixth day of incubation, 0. 25mg and 0. 50mg of 5-fluorouracil were injected into the chorioallantoic and the egg was broken for weighing the body weight and individual organs such as brain, heart, lung, liver and kidney on the day of 12th, 14th, 16th and 18th.
The effects of doses of the growth were analyzed by the analysis of variance method.
The formulas applied to determine the specific growth parameters : y=a+bt+ctz, y= bxa, and relative growth and the results thus obtained were summarized as follows:
when injected with 5-fluorouracil, the chick embryos were observed with remarkable .growth inhibition (P<0.005). The effect on the group injected with 0. 50mg was stronger than that in the group injected 0. 25mg.
Zimmanman¢¥s growth rate was increased with the development in the heart, lung, liver and kidney, on the other hand, It was decreased in the brain of 0.25mg and 0. 50mg injected group.
The specific growth rate of all organs of all groups showed tendency of decreased except the case of increased in the control groups of heart, lung and kidney and 0. 25mg injected groups of the heart and liver.
The coefficient of relative growth of the various organs to body weight in all group showed positive allometry while control and 0. 25mg injected groups of brain and liver sho¡þwed negative allometry.
The growth gradient of all organs of control and experimental group were on the eighteenth day of incubation except the brain and liver of 0. 25mg injected group and brain of 0. 50mg injected group which were on the twelfth day.
